Evidence Update

Current evidence

Nicotinamide mononucleotide (NMN), a different compound from nicotinamide riboside, was compared at 250 mg/day with placebo in a small trial of postmenopausal women with prediabetes. NMN improved a skeletal-muscle insulin-sensitivity readout in a small trial of prediabetic postmenopausal women, without establishing diabetes prevention or generalized metabolic rejuvenation.

What changed

The small prediabetic-women trial adds muscle insulin-sensitivity context, not a general functional-benefit claim.

What did not change

No diabetes-prevention, sarcopenia-prevention or aging-reversal claim is supported.

Why it matters

A tissue-specific physiological response does not establish prevention of diabetes or disability.

Main limitation

Hepatic/adipose insulin sensitivity and body-composition changes were not established; no clinical-incidence endpoint and limited sex/metabolic generalizability.

Next proof

Larger diverse trial measuring sustained clinical outcomes.

Sources

  1. Nicotinamide mononucleotide increases muscle insulin sensitivity in prediabetic women.

Source type primary_research_publication Publication date 2021

What this source establishes Reports muscle insulin-sensitivity physiology under nicotinamide mononucleotide in prediabetic postmenopausal women.

Important limitation The 25% muscle-uptake change is within the NMN arm, not a between-arm effect or diabetes-prevention result.
